To start navigating, any app needs to know your location. If your location is known, then you don't need to specify "route from". Only set "Route To" and press "Start".

Select a destination (using search, bookmarks, or tap on the map) and press Route To button. Then Start. Why it was hard for you to discover? In any other maps app you also need to select the destination first to start building a route/start navigation.

How to zoom in or zoom out with one hand in Organic Maps:
1. Tap quickly twice anywhere on the map, but don't lift up a finger after the 2nd tap.
2. Move your finger up or down to zoom in or zoom out.

Long Tap gesture: shows or hides the interface.

Navigation over an imported track is not ready yet. You may try to build a new route on top of the displayed track, and add necessary stops as a temporary workaround.
