Single-gene transcripts for subclinical tuberculosis: an individual participant data meta-analysis Single-gene transcripts are equivalent to multi-gene signatures for detection of subclinical tuberculosis, with consistent performance across settings. Check out our new paper in @lancetmicrobe.bsky.social

In this IPD-MA we show that single-gene transcripts are equivalent to multi-gene signatures to Dx subclinical TB

They perform consistently across settings + show potential clinical utility to stratify therapy

This was a fun analysis. We started out thinking blood RNA biomarkers of viral infection were likely to be correlated ++ as different dimensions of a common immune response (tinyurl.com/5ebfhsyj)

BUT the sampling resolution of human challenge helped us unravel the time course... IFI27 is unique ⬇️

2. Proprietary models eg Epic deterioration index. If the company are not willing/able to make the model reproducible, do we still want to see validation data?

I agree. 2 problematic areas on this:
1. Machine learning models where coefficients cannot readily be derived. Should we insist that the model object is shared in these cases? Does this create issues if the object includes the training data (can be tricky to fully remove)?

SARS-CoV-2 human challenge reveals biomarkers that discriminate early and late phases of respiratory viral infections - Nature Communications It’s not always clear whether blood biomarkers are differentially expressed in the time course of viral infections. In this SARS-CoV-2 human challenge study, the authors identify distinct single-gene ...
