We turned to population genetics for a solution. The resulting approach, MitoDrift, integrates Wright-Fisher drift dynamics with phylogeny inference via a hidden Markov tree model, enabling us to produce confidence-calibrated lineage trees.
Mitochondrial lineage tracing brought exciting new capabilities to single-cell genomics. However, a core problem remains: unlike classical lineage recorders, mtDNA mutations can drift and disappear as cells divide, preventing us from precisely decoding lineage history.
