I believe my lab did test before my time here different backbones of which a phosphorothiote ones were included. Will ask my PI what he thinks

Yes our lab is currently working on developing the concept of 6mer seed toxicity as a therapeutic.

We tested different toxic 6mer seeds varying only in 2-7 or 3-8pos embedded in a 19nt long ds RNA backbone. The sense strand was 2-O methylated to prevent RISC loading.
Excited to share my first publication from my postdoc at Northwestern University. It's been an interesting project and am glad to see it published in the Journal of Virology.
