For #FluorescenceFriday: a very thin slice of rat kidney was labeled with 8 primary antibodies whose signal was amplified with Aluora Spatial Amplification reagents. 🔬 on the EVOS S1000 Spatial Imaging system. >400 merged fields of view at 20x.

Under normal light fluorophore colors appear inverted: red dyes look blue, blue appear red, and green dyes are purple. Using an inverse LUT shows us this same palette on our microscopy sample. 🧪

LysoLight is ideally used in live cells; fixation is not recommended. Photostability is excellent, toxicity minimal, enabling long-term kinetic experiments.

Bright fluorescence is observed upon antigen-specific endosomal trafficking to the lysosome. Activation has been demonstrated with cathepsins B, D, & L. Inhibition is accomplished by blocking cathepsin, vATPase, or saturating cell-surface antigens with competing antibody.

The LysoLight family of dyes use a unique sensor that is activated by enzymatic cleavage specific to the lysosome, and therefore reports delivery of a protein or antibody to a functional lysosome. Minimal observable background fluorescence contributes to extraordinary sensitivity.

Several types of modern biotherapeutics require cellular uptake and lysosome delivery for efficacy. This includes ADCs, IgG-LNP, IgG-oligo, and of course extracellular targeted protein degraders like LYTACs. Existing fluorescence-based sensors report lysosomal delivery, but not catabolic activity.
