Together, these results highlight how ReactomeFI-powered network analysis combined with machine learning supports scalable predictive biomarker discovery in oncology.

Features from network topology and protein disorder were used to train Random Forest and XGBoost models, generating a Biomarker Probability Score (BPS) and identifying 2,084 candidate biomarkers, including high-confidence proteins such as LCK and ERK1.

Using Reactome Functional Interaction (ReactomeFI) as a primary pathway-informed network, the authors identified enriched three-node motifs containing oncologic drug targets and intrinsically disordered proteins.

In their November 2025 NPJ Systems Biology and Applications article, “MarkerPredict: predicting clinically relevant predictive biomarkers with machine learning”, Veres et al. introduce a network-based machine learning framework to identify predictive biomarkers for targeted cancer therapies.

Using functional assays and mutant viruses, the authors demonstrate that these #microRNAs directly target interferon receptors and core JAK/STAT signaling components, reducing antiviral gene activation during both early and latent infection.

The Journal of #Virology article "Inhibition of type I interferon signaling is a conserved function of gamma-herpesvirus-encoded microRNAs", examines how gamma-herpesviruses from non-human primates use microRNAs to suppress type I interferon signaling.
