ALL drugs are potentially unsafe. A rule of thumb in #geriatrics is that a new patient problem is likely from a new medication. It could be an interaction, it could be issues with drug metabolism. Many problems are better-address without medications at all. Prescribing is key to patient safety.
The story of Prontosil (an early antimicrobial) gives some idea: the inventor's daughter accidentally stabbed her arm with a knitting needle and the wound got infected. He was able to get her the drug and she was cured. Otherwise, her arm would have been amputated.

I don't know if everything has been linked so clearly, but I think it's a good way to look at it.
Not so random: the idea was to clear senescent cells, which release signals that inhibit regeneration (maybe including synthesis of prostaglandin E2). Senolytics are still a pretty active area of work, just didn't pan out in this case.
Millennial geriatrician here. There was a recent drug failure that also regenerated cartilage, targeting an aging mechanism. I hope they found a better target, but for now we'll have keep working out our quads 😕
