🏺 This matters because alternate peptide sequences can lead to inaccurate protein taxonomy and archaeological interpretations. The Demodifier is fast and open source, enabling easy detection of MISPs.

📊 The Demodifier was applied to a test dataset including 20 published studies of dental calculus and vessel residues, revealing that modification-induced sequence permutations occurred almost ten times more frequently than previously reported.

💻 The Demodifier simulates all possible modification-induced sequence permutations caused by deamidation (NQ) and N-terminal pyroglutamic acid formation, and assigns lowest common taxonomic ancestors via the Unipept API.

🧪 Protein modifications such as deamidation and N-terminal pyroglutamic acid formation can change peptide sequences and affect taxonomic assignment in palaeoproteomics.

New review paper by @mirandaevans.bsky.social
explores how #palaeoproteomicis is revolutionizing our understanding of how ancient people cooked 🏺
Future directions include developing minimally invasive sampling methods, better authentication techniques, and understanding preservation.
New paper out now📣
In this paper, now published in the Next Generation Archaeological Science special issue in JAS, I review the development and current state of the field of metaproteomic approaches to ancient foodways. 🏺🦷🥛🍲🧪 #palaeoproteomics
www.sciencedirect.com/science/arti...
3/3: In archaeology this is important because we are often trying to answer questions about which species were used by ancient people. The results show that this issue occurred 15% more frequently than currently reported.
www.biorxiv.org/content/10.1...
2/3: This is an issue in #palaeoproteomics because it can make it look like there is evidence for one taxon when in fact it could actually be a completely different one.
www.biorxiv.org/content/10.1...
1/3: Excited to share my new preprint, out today!
In it I share a tool I have created which screens for potentially spurious peptide taxonomies caused by post translational modifications.

Congratulations @isma-rgz.mastodon.social.ap.brid.gy on your first 1st-author paper.
Gets even better because it's with our favourite protein ever @matthewcollins.bsky.social @mirandaevans.bsky.social, eh!?
Apparently I talk with my hands. Thanks for having me to present on "The Proteomics of Pottery: what can dirty dishes tell us about ancient diet and cuisine?" at the Pudding Seminar, @Newnham_College!
