Interesting read. Aligns well with our findings (doi.org/10.1038/s414..., together with @haicovanattikum.bsky.social). We found DNA-PKcs inhibition to increase Cas9 break repair by single strand annealing (SSA), causing kilobase sized deletions. Exo1 depletion prevented this increase in SSA.
