🧐In a nutshell: CK1-mediated multisite phosphorylation of Dishevelled dramatically alters its bulk electrostatics, promoting DVL intramolecular coupling between the positively charged cleft of the DEP domain and the adjacent phosphorylated disordered region.
Excited to share our new preprint on bioRxiv. 🎉
We uncovered a Wnt-induced phospho-switch of Dishevelled. This intramolecular coupling is mutually exclusive with FZD engagement and functionally required for the Wnt/β-catenin signaling. 🧵

📌AH078 is a powerful tool for studying CK1 δ/ε biology in cells, enabling rapid depletion of both kinases—something challenging with KO systems due to cellular dependency on CK1 δ/ε activity!
👉 The PROTAC AH078 is characterized through in vitro and cellular assays, demonstrating on-target activity, selectivity, and a VHL-dependent mechanism of the CK1 degradation in cells.
🥳New paper from the lab!🍾 Excited to share our work with the Stefan Knapp lab (Goethe Uni, @thesgc.bsky.social & DKTK), published in J. Med. Chem. @pubs.acs.org. Here, we present the first degraders (PROTACs) targeting CK1 δ/ε—highly selective across the kinome & within the Casein kinase 1 family. 🧵
