I’m trying to make GR inducible protein expressed natively. I’ve made C & N terminal versions. Instead of using a genomic fragment with promoter to last aa, I used a 4kb promoter fragment and the CDS. I’m concerned that omitting intronic sequences will result in low expression. Thoughts?
