Capturing somatic mutations in single-cell probe-based assays (ie. 10X Flex) is a hard task, especially if you don't know *exactly* the mutations you want to capture, or if your target sequence is a mutational hotspot with many possible alternative alleles. Fibroblasts underpin immune-competent niches in the healthy and diseased CNS. Schematic representation of the topological organization of fibroblasts along the vascular tree and across meningeal and brain parenchymal compartments in the context of (A, B) homeostasis, (C, D) infection, (E) autoimmunity, and (F) sterile tissue injury. The provision of cytokines, chemokines, survival factors, and ECM components by local fibroblasts are indicated in each condition. The legend of depicted cell types and vasculature is provided. (A) In the homeostatic CNS, fibroblastic cells include mural cells, perivascular fibroblasts situated abluminal to mural cells, and meningeal fibroblasts. (B) In the steady-state condition, PDPN+ FRC-like cells are limited to DALTs positioned at the intersect of rostral-rhinal, olfactory, and superior sagittal sinuses. In (C) chronic dural infection and (E) autoimmunity, persistent inflammation is associated with TLS formation. TLS are underpinned by FRC-like cells and FDCs providing T and B cell chemoattractants, survival factors, and canonical markers of FRCs. (D) In infections of the brain parenchyma, similar chemokines and FRC markers are upregulated in the absence of TLS formation. (E) In the context of deep gray matter injury, perivascular fibroblasts of the Virchow-Robin space constitute a major source of scar-forming cells. In addition to profuse ECM deposition, scar-forming fibroblasts secrete myeloid cell survival factors and chemokines to drive early and late phase scarring, respectively.
