B cell-derived acetylcholine promotes liver regeneration by regulating Kupffer cell and hepatic CD8+ T cell function Liver regeneration (LR) is essential for recovery from acute trauma, cancer surgery, or transplantation. Fazel Modares et al. find that mouse LR after injury requires ChAT+ B cells synthesizing acetylcholine (ACh) and Kupffer cells and hepatic CD8+ T cells expressing α7 nicotinic ACh receptor (Chrna7). ChAT+ B cells drive Kupffer cell IL-6 secretion and reduce CD8+ T cell IFNγ.

nACh receptors engage a nice ChAT with the liver: B cells serving up regeneration Liver regeneration is a remarkable and unique biological process, orchestrated by an intricate cellular crosstalk of (non-)parenchymal, immune, and nerve cells. In this issue of Immunity, Modares et al. uncover the pivotal role of choline acetyltransferase (ChAT+)-expressing B cells as key orchestrators of liver regeneration.

UV1 vaccination in pembrolizumab-treated patients with recurrent or metastatic head and neck cancer: A randomized multicenter phase 2 trial The phase 2 FOCUS trial evaluated the hTERT vaccine UV1 plus pembrolizumab in recurrent/metastatic PD-L1+ head and neck cancer patients. UV1 was safe, did induce T cell responses, but it did not improve efficacy over pembrolizumab alone, suggesting limited benefit in this setting.

Deficiency of adenosine deaminase 2 skews adaptive immune repertoires towards specific sets of T and B cell receptors Adenosine deaminase 2 deficiency (DADA2) is a genetic disorder caused by biallelic hypomorphic or loss-of-function mutations in the ADA2 gene that encodes a protein deaminase regulating extracellular ...

We also developed a machine learning model that accurately distinguishes DADA2 patients from healthy individuals based solely on immune repertoire metrics, rather than sequence-level labels—shedding light on the unique immunogenetic signature of DADA2!

Excited to share our latest study on DADA2. We analyzed 52 patients, observing HGG, loss of T & B cell memory and persistent proinflammatory cytokines despite anti-TNF therapy. Notably, the B cell architecture was skewed toward IGHV4-34, a feature often seen in malignant B cells.

We observed phenotypic differences in T cells based on production methods - even without antigen exposure. In addition, transposon-based production appears to promote an exhaustion-like phenotype via RIG-I sensing of transposase-encoding mRNA.
