Intralysosomal pathogens differentially influence the proteolytic potential of their niche | Infection and Immunity Evading lysosomal degradation is of critical importance to successful intracellular pathogens. The lysosome, classically considered the waste disposal and recycling center of the cell, is a hostile en...

Coxiella & Leishmania both reside within a lysosome-like compartment in human cells, but appear to modulate the protease cohort in different ways. I’m pleased to share our latest story here, out this week in I&I: journals.asm.org/doi/10.1128/...

Engineered reactivity of a bacterial E1-like enzyme enables ATP-driven modification of protein and peptide C termini Nature Chemistry - In living systems, ATP provides an energetic driving force for protein synthesis and modification. Now, an engineered enzymatic tool has been developed for high-yield, ATP-driven...

Excited to share our latest: we engineered the reactivity of a bacterial E1-like enzyme for ATP-driven modification of C termini. Our tool mimics the logic of peptide bond formation in biology for precision modification of proteins in vitro. 🧪https://rdcu.be/ewN7C

Nuclear Roles for Canonically Lysosomal Proteases While the cysteine proteases legumain and cathepsins have traditionally been known as “lysosomal” proteases, there is increasing evidence to suggest that they also contribute to a wide range of ext...

Cutting the diagnosis journey for children born with rare genetic diseases Families can wait years for a diagnosis of a rare genetic disorder, but a new test can provide answers in days for a better understanding of the condition and potentially earlier treatment, finds new...

We're super proud to see our study showing utility of proteomics in ultra-rapid variant prioritisation for suspected mito and other rare diseases out in Genome Medicine (rdcu.be/endwE). Too many amazing collabs to thank, so here are the big ones @daniellahock.bsky.social @thorburnmito.bsky.social!

The phase IIb results from the multi center trail of our cathepsin probe (VGT-309/abenacianine) are out. The probe met its endpoints and resulted in significant clinical events in 45% of patients. Great work by Vergent Biosciences and Dr. Sunil Singhal (UPenn).

Coxiella burnetii manipulates the lysosomal protease cathepsin B to facilitate intracellular success - Nature Communications The zoonotic pathogen Coxiella burnetiiestablishes a unique intracellular niche within a lysosome-derived vacuole. We also show that Coxiella incites the extracellular secretion of lysosomal content in a T4SS independent manner.

A wonderfully enjoyable collaboration with @lauraem-lab.bsky.social & @hayleyjnewton.bsky.social labs (with significant mass spec guidance from @nickescott.bsky.social!)

Excited to share this story from my PhD, out today in Nature Communications! We show that Coxiella removes cathepsin B to reduce lysosomal hostility and promote bacterial success. This depends on a vacuole established by effector CvpB.

The ARC Board has proposed a major shake-up of the National Competitive Grants Program ▶️ www.arc.gov.au/engage-us/co...

I've only skimmed so far, but they propose reducing 13 grant schemes to 6, with intent & scope in the table👇

Submissions are being accepted in response until 13 April. Get to it!
