The MCLID mutations ultimately lead to impaired branching of hippocampal neurons in vivo (P7). Giving new insights into how these mutations can cause intellectual disabilities.
As part of this work, I created an optically controllable KIF11. With 405nm we can tie the hands and feet of KIF11 together, inhibiting it, resulting in more dynamic microtubules (seen below with more EB3 comets) and new dendritic growth.
These mutations were classified as “loss-of-function” but were never experimentally explored. By collaborating with the Diez Lab @TUD Dresden we showed these mutations differently affect KIF11/kinesin-5 function. If KIF11 is a bicycle one patient has a flat tire(Y82F) and the other has two unicycles
My paper is now online! We show how a mitotic kinesin supports microtubules in mature postmitotic neurons. When this motor is mutated in humans it can cause intellectual disabilities.
