All in all, RhoGAP15B locally suppresses RhoA–Myosin II at the leading edge, creating a relaxed cortical zone that stabilises protrusions while possibly helping to dictate the cycles of contractility/extension, thereby setting the pace for migration.
RhoA activity (measured with a FRET sensor) is increased with RhoGAP15B RNAi. While there is normal rear–front asymmetry in migrating border cell clusters, the loss of RhoGAP15B leads to elevated, spatially uniform RhoA activity, consistent with excessive cortical Myosin-driven contractility.
RhoGAP15B:GFP shows predominantly cortical localisation, with enrichment at the outer cortex. Within the leader cell, RhoGAP15B concentrates at the leading edge and is largely excluded from the nucleus and cytoplasm, consistent with a role in locally modulating RhoA–Myosin II at the migration front.
RhoGAP15B RNAi collapses leader protrusions, while RhoGAP15B OE stabilises and extends them, underscoring tight control protrusion dynamics.
How do migrating cell groups balance force and flexibility?
Our new preprint shows that RhoGAP15B downregulates RhoA–Myosin activity to stabilise protrusions and enable efficient collective migration in vivo.
