You may want to tell your agents to check out pydna github.com/pydna-group/.... It has functions for most cloning methods and primer design, and it tracks history. You can export your sequences as .gb files, but also .json that can be opened in the #OpenCloning website to see the cloning history

I've been working with the OpenCloning Assembler with the syntax of my Open Yeast System - both the Open Yeast Collection and Yeast Protein Expression Toolkit. It is a very powerful open approach for rapidly assembling constructs when you have a library of parts. Try it with your Golden Gate system.

New feature to help plan Modular Cloning (Golden Gate) experiments and streamline the otherwise confusing dance of overhang compatibility and vector hopping. Gonna find some time to try this out and compare to my current SnapGene workflow. Tools all day!
My favorite tool #Opencloning.org has just released a feature for assembling golden gate cloning fragments. I have not used it **yet**, but I've used the In-Fusion tool, and that ones has worked amazingly. Give it a try! I am sure @manu-lera.bsky.social will be happy to hear from your experience.

Working on a tool to define Golden Gate syntaxes for #OpenCloning. You define your syntax, and then you can drag-and-drop your plasmids in OpenCloning, they will be sorted into the right categories, and you can do combinatorial assembly. Reach out if you'd like to contribute:

I got a plasmid map from another lab as *.dna file 🧪. The SnapGene program can open it. I do not want to install their "free" viewer in my linux system. The solution: OpenCloning opencloning.org . With it, I loaded the .dna file and downloaded the sequence plus a nice map. Thanks OpenCloning team 🤩
