Exactly this happened to us. We expected to investigate (based on previous in vitro data) a conserved protein-lipid interaction in HIV, but what we actually discovered was much more exciting and groundbreaking. One still has to have an open mind to see that in the data.

Using any kind of angular priors (e.g. sample geometry based) helps a lot with the reconstruction and it is not restricted only to STA. :)
Could you specify the gain in resolution?
