Team: This has been an extraordinary effort from the whole team but especially by co-first authors Florian Kromm, Søren Borcher Møller who overcame countless obstacles along the way. @au.dk @thomastorring.bsky.social

A few highlights:
-11 steps in the longest linear sequence to lasalocid acid A. Five building blocks - four interesting C-C fragment couplings!
-Pivotal Pd/ZnF₂-mediated C-C coupling reaction affords the attached ring product in high yield. Broad substrate compatibility - multiple diastereomers.

Thrilled to share our new paper just published in @acsjacs.bsky.social: a modular, 11‑step total synthesis of lasalocid acid A—enabled by a new Pd‑catalyzed C-glycosylation reaction to construct C(sp³)–C(sp³) attached‑rings.

Synthesis and functionalization of vinyl sulfonimidamides and their potential as electrophilic warheads Covalent inhibitor design is dominated by the use of electrophilic acrylamide warheads. One limitation of acrylamides is that there are limited opportunities to modify their electrophilicity, and henc...

Great to see Ivan and Charlie's work on the synthesis and reactivity of vinyl sulfonimidamides out in @chemicalscience.rsc.org Here they show that reactivity either above or below that of the corresponding acrylamide can be achieved. @oxfordchemistry.bsky.social

Our new route represents a dramatic shortening compared to classic routes to this compound, which was strongly enabled by development of a new transformation to directly couple the challenging attached oxacyclic rings. We are further excited about the generality of this C(sp3)-C(sp3) coupling method

Delighted to share our newest pre-print. Here, we describe a very short synthesis (11 steps, LLS) of the polyether antibiotic lasalocid acid A.

Ten simple rules for developing good reading habits during graduate school and beyond

To me, the most important are:
Read often, read broadly (incl. older papers and outside your field), and learn to read some papers in detail and others more superficially (and quickly)

Formation of mono- and dual-labelled antibody fragment conjugates via reversible site-selective disulfide modification and proximity induced lysine reactivity Many protein bioconjugation strategies focus on the modification of lysine residues owing to the nucleophilicity of their amine side-chain, the generally high abundance of lysine residues on a protein...

Here is a very interesting new paper from the Baker and Chudasama groups at University College London. A clever example of leveraging reversible disulfide rebridging chemistry to achieve selective lysine modification (via proximity) of antibody-fragments.

It is great to have @aekiib.bsky.social from the group of @tbpoulsen.bsky.social as visiting PhD student in the lab. His stay in our lab allows us to exchange a lot of thoughts and technologies between the labs and learn so much from each other. #ChemBio onlinelibrary.wiley....

This looks like an interesting application of the oxSTEF probes developed in the Poulsen lab before. Good example of developing interesting chemistry/chemical biology and then asking the right biological questions.

Interesting ChemRxiv preprint by the group of @tbpoulsen.bsky.social with first author @esbensv.bsky.social. They used OxSTEF probes to study proteins with N-terminal cysteines in the proteome and used the method to specifically investigate difference in normoxia and hypoxia. doi.org/10.26434/che...
