LLOMe has long been used to study lysosomal damage, yet how it works has remained a mystery.
Using cryo-electron tomography, we show it forms amyloid structures inside lysosomes that mechanically rupture membranes – revealing a new paradigm for lysosomal failure.

Good guess, but Chlamy lacks 'canonical vaults'. My speculation for the lightbulb complex would be one of the five name-less Chlamy SPFH proteins (www.ebi.ac.uk/interpro/ent...), or maybe Prohibitins, though they’re usually IMM-localized in other organisms.

It’s out! Using cryo-ET in Dicty cells, we take a fresh in situ look at vaults. Surprisingly, we uncover vaults associated with ER and NE membranes, and find that many vaults enclose ribosomes in defined orientations, opening new avenues to their cellular function! www.biorxiv.org/lookup/conte...
