Congratulations to Lukas @pekarekl.bsky.social for winning the poster prize at the DGfB Molecular Biophysics meeting in Hünfeld last week 🏆 - it was a fantastic meeting with lots of interesting talks & great to catch up with colleagues and sad that we missed out on many of our Israeli colleagues.
... and finally saw that the survival fraction goes down 4-fold in presence of the peptide. Yay - this sounds like another great new adventure ahead. Great work by Kate and Pilar, 🙏 support by @tudresden.bsky.social and BMFTR. Take a look at bioRxiv to find out more 🔎: doi.org/10.64898/202... 6/6
This was a new adventure for us to develop an assay to stress bacteria 🧫 and test how good they adapt to antibiotic stress. We stress bacteria 🦠 with e.g. Ciprofloxacin and see if they adapt via a provided mobile integron system ± peptide. We determine many factors using also microscopy… 5/
…modify single amino acids to gain or lose function. It is dose dependent and works with a peptide as short as 7 aa. But wait 🫷- does it also work in bacteria? 4/
… use this to find a small molecule, which inhibits tail binding and thus destabilises the complex and reduces the ability to adapt to antibiotic stress in bacteria? Yes, and Kate 👩🔬 came up with a few different peptides … and we can control destabilisation … 3/
DNA elements were important for mechanics, but also protein mutations decreased recombination significantly. In particular the C-terminal extensions (the little pig-like ;) tails linking IntI1) stabilised the complex significantly and truncation reduced recombination. Hm💭 - could we maybe … 2/
Our newest bioRxiv on the bacterial integron is out (link below). 🧑🔬
How it started: Rising #AMR are a challenge for health. A key player is the bacterial integron. Do you remember its recombination efficiency 🧬🧬 correlated with the mechanical stability 💪 of a recombinase-DNA complex? 1/6
