This work was also our first time doing membrane localization (way to go Cassidy)! And so proud of Bella’s gorgeous Paenibacillus tree showing the distribution of the transmembrane containing Spo0B! Also it’s the lab’s first paper without ribosomes 🫨, but don’t worry - the ribosomes made the Spo0B!

In this study, we highlight the importance of the base composition of the first codons for protein expression. An increased abundance of adenine (A) and a decrease in guanine (G) enhance expression and improve mRNA stability. The mediator of this effect is the ribosome.

The second method does the same but also
optimizes all other codons using the 44 best codons identified in our
previously published efficiency codon metric (doi.org/10.1038/natu...). The last method (AG29G) follows a similar
approach but utilizes only the 29 best codons.
The first
optimization method (AGhead) focuses solely on the first codons to increase the
ratio of adenosine to guanosine.
The manuscript comes with a web server, opt.williams.edu, that allows for the prediction of expression for
a given DNA sequence and proposes three types of optimization.
I am very happy to present our new manuscript in collaboration with the Aalbet and Hunt Labs on DNA sequence optimization for high expression in Escherichia coli as well as in in vitro translation assays (doi.org/10.1016/j.jm...)
