This method not only saves time, but is also economical, more efficient and mild in isolation conditions, preserving translation complexes, ribosome associated proteins and enables isolation of ribosomes for structural (cryo-EM) and functional studies. More on RAPPL uses still to come...
Our RAPPL purification of ribosomes and associated material is out. I am really proud of this study in collaboration primary with Pavlovic-Djuranovic lab, as well as Jovanovic and Hashem Labs. This method aims to replace 60 years old purification of ribosomes by sucrose cushions

Our collaboration with Tony Mustoe's lab us out today. MPRA libraries and possible cryptic splicing in MPRA reporters, or reporters in general. The additional novelty here is that such events are controlled or influenced by AU-rich sequences.

My suggestion to everyone is to keep some of your most valuable cell lines and back-ups in your collaborators freezers or at least in different buildings or towns.
I should point out that this was a valuable collection of mutant cell lines as well as CRISPR/Cas9 engineered lines with endogenously tagged-RBPs, translation factors, ribosomal proteins and other proteins.
3 years after terrible loss (two -80C freezers and all back-ups) of almost all of our collection of regular and mutant human, Drosophila, E. Coli and Plasmodium cells, we are still not finished with insurance.
