Very excited to announce that my first, first author paper, is now on BioRxiv! In this paper we unambiguously show #COPII coated vesicles in unperturbed human cells for the very first time!!

Make my day and check it out!

This revolutionised our segmentation of coated membranes.

Membrain V1 didn't recognise coated membranes - lots of manual editing was needed. Membrain V2 - beautiful segmentation, chefs kiss!

Yes! we reproducibly get ~150nm thin lamella with very few failures and regularly run sessions for ~48hrs.

We have the Aquilos2 which is fab!

Our only complaint is the iFLM filter cubes are optimised for room temperature but the spectra of a number of fluorophores changes at cryo and so their peak excitation and emission wavelengths are filtered out leading to poor signal at cryo vs RT

🦠🧠 MemBrain update! 🧠🦠
We’ve updated our preprint! It now covers the full MemBrain v2 pipeline for end-to-end membrane analysis in #CryoET: segmentation, particle picking, and spatial statistics.

Any guesses of whats going on here? #TeamTomo

Prize? idk... bragging rights? The feeling of superiority?

I'll collect a high mag tomo tomorrow, so the answer may actually be revealed! (mysteries mysteries mysteries).

This scheme shows the cargo adaptor FHF in the middle with arrows pointing to distinct microtubule and actin-based motors (dynein-dynactin, KIFC1, KIF1C and myosins V, IX and X as well as Tropomyosin 1). It also shows a link to Rab5 marked early endosomes. The hypothesis is that FHF coordinates multi motor binding at the early stages of endocytosis. The PhD project would address the hierarchy of motor binding and how these motors coordinate to allow the newly formed vesicle to transition from the actin to MT cytoskeletal network.

If you’re interested in how molecular motors coordinate to power intracellular trafficking events (e.g. endocytosis), I’ve got a PhD position available to study this at vibrant Bristol uni. In this project, we will capture highly dynamic events by #cryoEM, single molecule imaging & cell biology. 🔬❄️🧪
