Genome-scale perturb-seq in primary human CD4+ T cells maps context-specific regulators of T cell programs and human immune traits Gene regulatory networks encode the fundamental logic of cellular functions, but systematic network mapping remains challenging, especially in cell states relevant to human biology and disease. Here, ...

Today's the print release of a #RibackLab and #GoodellLab collaboration— we’re excited to share the visual story behind the science, including the dramatic artwork that inspired how we thought about aberrant function of NPM1c - which occurs in about 1 in 3 adult acute myeloid leukemias (#AML).

Big news! Our team developed Neuro-PERC, a non-viral CRISPR delivery platform for genome editing of the brain. In our new preprint - bit.ly/NeuroPERC_pre - we report efficient editing in multiple models & species, including field-first results in large animals (pigs) 🧵👇

Super cool! Congrats! Have you looked into immune responses towards Cas proteins and (Neuro-)PERC peptides?

Workflow of EBV-seq

Workflow of EBV-seq

Adapting the 10x workflow to detect EBV reads, Younis et al show that EBV-transcribing B cells are increased in SLE patients, especially among CD27+CD21low B cells. They have autoreactive BCRs and are more poised for antigen-presentation.

CD8 T cells produce IFNg and CXCL13 in TLS

CD8 T cells produce IFNg and CXCL13 in TLS

What mechanisms drive checkpoint inhibitor-associated interstitial nephritis (ICI-AIN)?
In our preprint, we identify CD8 T cells as a central player, inducing a proinflammatory myeloid cell state by IFNg and structuring lymphoid aggregates by CXCL13.

(1/10) How do diverse leukemia mutations converge on the same molecular program? In #RibackLab first manuscript @cp-cell.bsky.social, collaboration with @goodell-lab.bsky.social shows that disparate mutations rewire shared protein networks to form nuclear condensates called C-bodies.
