(5/5) Incorporating the engineered HMAs domains into TKP receptors, we demonstrate the engineered receptors have expanded recognition profiles in cereal protoplast assays.
(4/5) The effector binding interfaces on the HMA domains of Rmo2 and Rwt7 are spatially distinct. We engineered both TKP-HMAs to gain binding to non-cognate effectors in vitro and in plant cells.
(3/5) Based on these structures, we mutated the effectors disrupting TKP-HMA binding. This prevents TKP recognition responses in infection assays confirming the role of the TKP-HMA as an integrated effector bait domain.
(2/5) We experimentally solved the co-structures of TKP-HMA domains from Rmo2 and Rwt7 with cognate effectors, PBY2 and PWT7, respectively and show they bind tightly and specifically to the effectors that they recognise.
(1/5) Rmo2 and Rwt7 are TKPs that mediate immunity to blast disease in barley and wheat, respectively. Both receptors contain an integrated HMA domain. Check out the preprint from our collaborators:
