(4/5) The effector binding interfaces on the HMA domains of Rmo2 and Rwt7 are spatially distinct. We engineered both TKP-HMAs to gain binding to non-cognate effectors in vitro and in plant cells.

(3/5) Based on these structures, we mutated the effectors disrupting TKP-HMA binding. This prevents TKP recognition responses in infection assays confirming the role of the TKP-HMA as an integrated effector bait domain.
