Our comprehensive analysis of human iNKT cells from multiple immunologically relevant hematologic tissues provides critical insights into their transcriptional heterogeneity and an invaluable resource for future research and development of iNKT-based therapies to optimize their safety & efficacy.
We saw 2 transcriptionally distinct CD8+ iNKT cell populations, one naïve/precursor-like & one Th1/17/NK-like, & several distinct CD4+ populations, including a Th1/17/NK-like one. These findings suggest that CD4 & 8 expression (or lack thereof) does not always correlate with functional propensity.
A combined Th1/17 transcriptional signature was detected in some human iNKT cells (verified by co-expression of Tbet & RORgt protein)--similar to human MAIT cells, yet different than mouse iNKT cells. We further uncover unique transcription factor dynamics and identify TEMRA-like iNKT cells.
