High efficiency CRISPR knock-in demonstrates that TCF1 is insufficient to reverse T cell exhaustion - Nature Communications Exhaustion is a functional state that hampers anti-cancer and antiviral CD8 T cell activity, and is preceded by a stem-like state, maintained by the transcription factor TCF1. Here authors develop mou...

TCF1 is a master regulator of T cell stemness – but can it make exhausted cells go backwards?

We setup a CRISPR KI system to ask this question, and the answer was unexpected.

γδ T cell-derived IL-4 initiates CD8+ T cell immunity - Nature Immunology The authors show how Vγ1+ γδ T cells produce IL-4 to drive early CD8+ T cell and dendritic cell responses to malaria infection in mice.

Ever wondered why gamma delta T cells are required for immunity to liver-stage malaria?

Medical Research Matters

Right now, the Government is starving medical research of full funding, even as it claims that science and research are national priorities. If our leaders truly value science as much as they say, then they must back their words with action—and fund it properly.

Microbiota-derived butyrate promotes a FOXO1-induced stemness program and preserves CD8+ T cell immunity against melanoma The gut microbiome impacts immunity at various levels, including response to immunotherapy. Bachem et al. find that metabolic pathways shared across different microbiota constituents, including SCFA synthesis, modulate stem-like tumor-specific CD127+CD8+ T cells in the tumor-draining lymph node and impact control of melanoma.

Trump 2.0: an assault on science anywhere is an assault on science everywhere US President Donald Trump is taking a wrecking ball to science and to international institutions. The global research community must take a stand against these attacks.

Inexplicably, US administration has launched an unprecedented assault on science, on research institutions & on vital international organizations & initiatives.

How could this work?

IL-1 cytokines expand ID3+ stem-like T cells in vitro, boosting their ability to sustain T cell responses in chronic infections & cancer.

This could pave the way for next-gen T cell therapies with enhanced longevity & efficacy. Exciting possibilities ahead!

ID3+ T cells in acute settings show unique stem-like features, including the ability to sustain long-term immunity and exclusively form #Tpex cells in chronic infection & cancer.

Could this improve durability in T cell therapies? We think so! 🧬

#Tpex cells are key to sustaining T cell responses to chronic infection & cancer. Can we learn from #Tpex cells, and harness stem-like T cells in acute infection to improve chronic immune responses?

Yes! T cells identified in acute LCMV expressing ID3 and LY108 shape durable therapies
