For deeper fluorophores, #HILO is a great choice, with the laser directed at the sample in an angle not as acute as in TIRF, while maintaining a low background. In #Epifluorescence (or #Brightfield), the laser passes directly upwards through the sample, maximizing the amount of illumination.
