Finally, we illustrate the relevance of pTHG microscopy for characterizing myelin distribution at different scales in fixed mouse and human brain tissue.
Fun fact for multiphoton microscopy geeks: aberrations caused by vertical micro-interfaces appear to be a significant contrast mechanism in polarized THG imaging. 😎
We show that axons exhibit complex pTHG profiles that depend on axon morphology and can't be explained by classical models. We establish a numerical strategy to relate pTHG profiles to axon diameter and myelin thickness, taking into account myelin-induced optical aberrations.
Our study presents a systematic characterization of polarized THG (pTHG) signals from individual axons, enabling the derivation of myelin scores in various systems. 🐭🐟
We confirm the sensitivity of THG for myelin in mouse tissue and detect early axon development in zebrafish.
