Hi all!
We developed CAD-C to map chromatin organization at high res in yeast. It revealed an unexpected tight alignment: nucleosomes covering the same sequence tend to be associated across sister chromatids.

Such « post-lesion » hybrids can be detected in yeast and human cells, yet they do not detectably impact genome stability, as opposed to « pre-lesion » hybrids - a revisited classification? (5/8)
For example, focusing on Okazaki fragments (OF) processing, we found that the accumulation of unprocessed flaps or unligated OF termini is the source of hybrid formation 🧬 (4/8)
We identified several situations where DNA:RNA hybrids are associated with genetic instability – strikingly, in most cases, DNA lesions appear to precede hybrid formation 🔃 (3/8)
