You are right, it's not a BONCAT problem per se, it only becomes a problem for the FACS part of BONCAT-FACS. The dyes reacting with the peat was not a major problem, it was quenching and autofluorescence that made it tricky. We tested different azide-dyes and ended up using Cy5 as that worked best.
True, but having worked with peat for years we somehow expected problems on the fluorescence side. It was getting clean enough cell preparations for FACS sorting that proved to be rather challenging. But we got it sorted in the end.
Depending on the system they work in people will likely face quite different challenges. We for example were doing quite ok with labelling etc., but working with peat it has been extremely challenging getting the cells separated from the background fluff (which has basically the same density).
Great idea, this would be quite helpful for many! Even though I have to say that your group's papers on the topic are quite detailed already. I think Microbiology Spectrum and Msystems would be quite fitting, at least they also support publication of methods/protocols without data.
