New preprint from the lab! We discovered a transient fluidization in the basal region of human forebrains by tracking microdroplets in cerebral organoids. This “basal fluidization”, absent in gorilla and mouse, may contribute to greater surface expansion in human forebrains

The goal of a PhD is not to learn some facts or read a few papers or learn a bunch of techniques. The goal of a PhD is to learn independence, problem solving, how to finish things you start, resilience, & gain the ability to adapt & think creatively. Learning these things is hard.

Overall, this study comprehensively characterizes the epigenetic state of specific neural cell types and highlights a novel role for H3K27me3 in regulating the ECM composition in the human developing neocortex. (10/11)

Intriguingly, pharmacological PRC2 inhibition in human cortical organoids induces increased production of the ECM components Syndecan 1 and Laminin alpha 1. (9/11)

Our findings are especially interesting considering previous results from the Hippenmeyer lab, where PRC2 knockout in individual cells of the mouse cortex suggested a non-cell autonomous mechanism for the regulation of NPC fate decisions by PRC2. (8/11)

Our cell type-specific profiling of the genome-wide H3K27me3 profile not only identified neuronal differentiation, but also extra-cellular matrix (ECM) genes as targets of PRC2. (7/11)

Whereas a recent study from the Treutlein lab and @epign.bsky.social revealed a role of PRC2 in regulating the exit from pluripotency, we now dissect the role of PRC2 within the neural lineage after the initial cell fate commitment. (6/11)

We found that loss of H3K27me3 in human cortical organoids induces a shift of neural progenitor cell proliferation towards differentiation that results in reduced organoid size. (5/11)

Here, we performed pharmacological inhibition of PRC2 in human cortical organoids from week 3 to 8, leading to a near complete loss of H3K27me3, and studied NPC behaviour. (4/11)

An epigenetic barrier sets the timing of human neuronal maturation - Nature The slow maturation of human neurons is regulated by epigenetic modification in nascent neurons, mediated by EZH2, EHMT1, EHMT2 and DOT1L.

These results beautifully align with a recent study from the Studer lab where PRC2 was shown to contribute to an epigenetic barrier that mediates the protracted maturation of human neurons compared to other species. (3/11)

We apply an innovative, highly multiplexed approach, Epi-CyTOF, to study > 30 epigenetic modifications at single-cell level in the human developing neocortex. We identify H3K27me3, deposited by PRC2, as the modification with the strongest cell type-specific enrichment. (2/11)
