We believe these findings provide important insights into how T cells adapt across organs and how tissue cues shape both their function and clonal expansion. We also hope that TRACK mice will be a valuable tool for the scientific community studying T cell dynamics in health and disease.
Interestingly, early clonal segregation during infection evolved into greater inter-organ sharing, leading to clonal equilibrium over time, likely driven by retrograde migration previously reported in CD8 T cells. Additionally, TCR specificity converged on a few viral proteins at the memory stage.
During viral infection, different tissues, including secondary lymphoid organs, not only shape the transcriptional profile of CD4 T cells but also control the expansion of specific clones, likely driven by TCR specificity and each tissue’s unique antigen landscape.
The novel TRACK mice we developed allow us to carefully follow recently activated, antigen-agnostic polyclonal CD4 T cells during perturbations (controlled by tamoxifen) and determine how they are clonally selected and transcriptionally defined across organs.
