🧬 #PKA has long been considered essential for hippocampus-dependent long-term memory, but deleterious for #WM.
We demonstrate that PKA activation in the striatum is beneficial for working memory capacity. This calls for targeted, region-specific PKA modulators — intelligent PKA agents.

✅ We confirm that low doses improve memory in a schizophrenia mouse model, using the same dose range previously applied by #PatriciaGoldman-Rakic in monkeys (unlike the much higher doses in recent rodent studies).

✔️ High-dose recruits PKA in the cortex — again, not novel — but:

Cortical PKA activation recruits parvalbumin interneurons in the #striatum, shutting down the striatum, thus impairing memory.
Thus: a fronto-striatal circuit mechanism for the dose-dependent effects of DA drugs on memory.

🔍 So, what’s new?

✔️ Low-dose D1 stimulation expands MC via #PKA activation in the striatum.
We show that striatal PKA activation is both necessary and sufficient to enhance memory capacity.

📢 New paper!! we show that memory capacity (MC) in mice can be expanded from 6 to 8 objects using low doses of a D1 receptor agonist. Higher doses? Paradoxically, they reduce MC.
