My personal rule is: always be kind, understanding, and willing to listen to your female colleagues. Whatever they have achieved, in most cases it is more than deserved and they have something to teach you!

⚖️ Takeaway?
The good and the bad of dopamine all depend on the dose — and the circuit engaged.

🧪 Our #phosphoproteomic data reveal that low vs. high doses of D1 agonist trigger completely different signaling profiles.
It’s as if we were administering two entirely different drugs — astonishing!

🧬 #PKA has long been considered essential for hippocampus-dependent long-term memory, but deleterious for #WM.
We demonstrate that PKA activation in the striatum is beneficial for working memory capacity. This calls for targeted, region-specific PKA modulators — intelligent PKA agents.

🧠💡 Why does this matter?

✅ We confirm that low doses improve memory in a schizophrenia mouse model, using the same dose range previously applied by #PatriciaGoldman-Rakic in monkeys (unlike the much higher doses in recent rodent studies).

✔️ High-dose recruits PKA in the cortex — again, not novel — but:

Cortical PKA activation recruits parvalbumin interneurons in the #striatum, shutting down the striatum, thus impairing memory.
Thus: a fronto-striatal circuit mechanism for the dose-dependent effects of DA drugs on memory.

This inverted-U dose-response curve is well-known in the literature — nothing new here.

🔍 So, what’s new?

✔️ Low-dose D1 stimulation expands MC via #PKA activation in the striatum.
We show that striatal PKA activation is both necessary and sufficient to enhance memory capacity.

🧠💥 For #Dopamine (DA) & #Memory lovers!

📢 New paper!! we show that memory capacity (MC) in mice can be expanded from 6 to 8 objects using low doses of a D1 receptor agonist. Higher doses? Paradoxically, they reduce MC.
