Very excited to share our latest work in #MolCell showing NPM1 stabilizing the association of nucleolus associated domains (NADs) and recruiting G9a to establish their repressive chromatin states. www.sciencedirect.com/science/arti...

The nucleolar granular component mediates genome-nucleolus interactions and establishes their repressive chromatin states Gupta et al. showed that NPM1, a nucleolar granular component factor, mediates NAD association with the nucleolus. NPM1 binds NADs and recruits histone methyltransferase G9a to establish H3K9me2 repressive chromatin, revealing that the nucleolus functions not only as a scaffold for NADs but also actively contributes to their chromatin repression.

Single-molecule analysis reveals the mechanism of chromatin ubiquitylation by variant PRC1 complexes Single-molecule experiments show that active conformation formation controls chromatin ubiquitylation kinetics by variant PRC1.

Our new study of chromatin ubiquitylation by variant PRC1 on the single-molecule scale: We visualize directly how vPRC1 ubiquitylates neighboring nucleosomes during a single binding event, showing a potential mechanism how H2Aub domains are established.

Reconfiguration of genome-lamina interactions marks the commissioning of limb cell-fates Diverse forms of heterochromatin block inappropriate transcription and safeguard differentiation and cell identity. Yet, how and when heterochromatin is reconfigured to facilitate changes in cell-fate...

Very excited to share our latest work revealing a prominent role for LADs in commisioning cell fates in mouse organ & tissue development, particularly of the limb. Amazing work by Isabel and Samy together with @stemundi.bsky.social & @drmrobson.bsky.social

ZNF574 is a quality control factor for defective ribosome biogenesis intermediates Without surveillance, errors in ribosome assembly can lead to the buildup of defective intermediates that impair protein production. Akers et al. developed a system to model faulty ribosomes in human cells and discovered that the protein ZNF574 targets these intermediates for degradation. Loss of quality control results in defective ribosome buildup that jeopardizes cellular homeostasis and organismal health.
