Channel 4's recent docudrama Dirty Business highlighted the shocking scale of pollution of England's rivers and seas. It showed devastating health harms linked to releases of untreated sewage, along with statistics and personal stories. In 2024, water companies in England made nearly a million illegal sewage dumps. Surfers Against Sewage reported that around 1500 people contacted it about illness caused by swimming in sewage contaminated water in 2023.

Elraglusib and chemotherapy in metastatic pancreatic ductal adenocarcinoma: a randomized controlled phase 2 trial - Nature Medicine In this phase 2 trial, combination treatment with elraglusib, a cell-permeable ATP-competitive inhibitor of glycogen synthase kinase-3β, and gemcitabine plus nab-paclitaxel (GnP), in patients with previously untreated metastatic pancreatic ductal adenocarcinoma led to prolonged overall survival compared with GnP only.

Elraglusib with chemotherapy boosts survival in metastatic pancreatic cancer, extending median overall survival by 2.9 months compared to chemotherapy alone.

Here we identify farnesyl pyrophosphate (FPP), a mevalonate pathway metabolic intermediate1, as an endogenous alarmin that enhances IgG antibody responses through keratinocyte-derived IL-6 and CCL20. This signalling axis potentiates the differentiation of T follicular helper cells and migratory dendritic cells2,3. FPP accumulates within keratinocytes after infection or ultraviolet irradiation through the activation of the mevalonate pathway mediated by the unfolded protein response–SREBF pathway, amplifying germinal centre (GC) responses in draining lymph nodes. Mechanistically, accumulated FPP in the cytosol engages transient receptor potential vanilloid 3 (TRPV3) by binding to its intracellular domains, inducing Ca2+ influx that subsequently activates the calmodulin–calcineurin–NFAT and PYK2–RAS–ERK pathways to enhance IL-6 and CCL20 production. Pre-splicing conformation and stepwise circularization of a group I intron in Azoarcus pre-tRNA | Nature Communications Group I introns are catalytic RNAs capable of self-splicing, yet structural insights into full-length precursor RNAs and post-splicing circularization have been limited. Here, we present cryo-EM structures of the Azoarcus pre-tRNAIle system across key catalytic stages, including the full-length precursor in Pre-1S conformations, the linear intron, and the circular introns, at 2.8-3.3 Å resolution. These structures reveal a preformed P1 helix, register shifts during splicing, and a conformational flip of G37 that stabilizes the circularization site. Biochemical analyses confirm a two-step circularization mechanism, generating a secondary circular product via an alternative ligation site. Together, our results provide an atomic-level view of a group I intron system through splicing and circularization. This work uncovers structural principles governing RNA conformational dynamics, catalysis, and circular RNA formation, with broad implications for ribozyme engineering. Cryo-EM structures
