Here we show that #nitisinone kills insecticide-resistant mosquitoes via cuticular uptake. Great work Zach Stavrou-Dowd, Lee Haines, & LITE. Thanks @gatesfoundation.bsky.social & @RoseTrees Trust @notredame.bsky.social @lstmnews.bsky.social parasitesandvectors.biomedcentral.com/articles/10....
New paper out in Parasites & Vectors!
We show that nitisinone, a repurposed β-triketone, kills Anopheles gambiae via tarsal contact, even in pyrethroid-resistant strains.

#Nitisinone already has regulatory approval for treating certain rare, inherited diseases. It works by blocking the production of a specific protein, which leads to reduced toxic disease byproducts in the human body. But when mosquitoes drink blood with nitisinone, they quickly die.
The drug in question is #nitisinone, and a proof-of-concept study led by a team from the Liverpool School of Tropical Medicine in the UK found that it could be deadly to mosquitoes at a low dose in human blood. The insects died within 12 hours.
The drug #nitisinone, already approved in the US to treat metabolic disorders, can make human blood toxic to mosquitoes.
Female Anopheles gambiae #mosquitoes died within 12 hrs of consuming blood taken from 3 people who regularly take nitisinone.

🦟 "What if human blood were toxic to #mosquitoes? A drug can make it so.
A single dose of the drug #nitisinone could render a person’s blood lethal to #Anopheles mosquitoes for five days, modelling suggests." @nature.com

Glad this is finally out. Using pharmacokinetic modeling we show that #nitisinone, an FDA-approved drug 4 #Tyrosinemia & #Alkaptonuria, kills #Anopheles mosquitoes, outperforming #Ivermectin. Grateful to all primary author’s & fantastic collaborators!

In this work we show that #nitisinone kills both susceptible and insecticide-resistant Anopheles gambiae mosquitoes through cuticular absorption. Our findings support development of a new type of insecticides that specifically target the mosquito Tyr pathway without affecting insect pollinators.
Nitisinone: Warnings & Precautions.
Nitisinone use has been linked to low platelet counts, often known as severe thrombocytopenia and leukopenia.
