Top: Overview of the scRNA-Seq experiment; WT young (2–3 months of age), WT middle-aged (8–12 months of age), MycTg middle-aged, WT aged (15–18 months of age), and iMycTg aged-E (enlarged) (15–18 months of age) mice were infected with 10 cysts of T. gondii. T-cells were isolated from the spleen on day 12 after infection. Middle: A UMAP of the annotated CD4 and CD8 T-cell populations. Bottom: Bar plot showing the abundance of the annotated CD4 and CD8 T-cell populations.

Age-related thymic decline impairs #Tcell #immunity, increasing infection risk in the #elderly. This study shows that enhancing #thymic function in aged mice restores naïve T cells and improves immune responses, thus supporting thymic regeneration in aging @plosbiology.org 🧪 plos.io/46yMgW3

I’m pleased to share that I’ve been awarded an Independent Investigator Grant from @RallyFoundation for Childhood Cancer Research to support our research on regenerating #thymic function.

Grateful to be part of this mission.

Let’s #RallyOn and stay #GOLDSTRONG for every child and family affected.

ONLINE COVER: Lymph Nodes Get a Remodel. Fibroblastic reticular cells (FRCs) are stromal cells that coordinate structural remodeling and adaptive immune responses in the lymph node. Using single-cell transcriptomics and high-resolution microscopy, Lütge et al. comprehensively characterized the stromal cell landscape of human lymph nodes during quiescence and inflammation. Inflammation drove lymph node remodeling including the expansion of peptidase inhibitor 16 (PI16)+ FRCs in macrophage-rich perivascular niches. This month’s cover image shows an inflamed human lymph node that has undergone expansion of podoplanin+ FRCs (purple) near alpha smooth muscle actin (ACTA2)+ vascular smooth muscle cells (green) to support immune cell infiltration and BCL6+ germinal center B cell (orange) activity.

Phase II Parallel Arm Study of Sacituzumab Govitecan-Hziy in Patients with Advanced Thymoma or Thymic Carcinoma Thymic epithelial tumors (TETs), including thymoma and thymic carcinoma, are rare thoracic tumors of the anterior mediastinum. For those with advanced disease, platinum-based chemotherapy is used as first-line treatment. However, there is no standard regimen established for TET at progression after initial therapy, and treatment options for advanced/recurrent TETs are limited. Trop-2, a transmembrane glycoprotein, is overexpressed in solid tumors including thymomas and thymic carcinomas. Sacituzumab govitecan-hziy, a Trop-2-directed antibody-drug conjugate, has shown efficacy and safety in several tumors including breast cancer.

Trials in progress at Clinical Lung Cancer describes ongoing phase II study of sacituzumab govitecan (TROP2 ADC) in #thymic carcinoma or thymoma. Led by Drs. @chulkimmd.bsky.social @jennifermarksmd.bsky.social. Open to enrollment now! #OncSky

CD8 T cell tolerance results from eviction of immature autoreactive cells from the thymus Thymic eviction explains CD8 T cell tolerance and why autoreactive CD8 T cells appear in the periphery but not the thymus.

#TCR #crossreactivity "Premature #thymic #eviction is the basis for #CD8Tcell tolerance and is the mechanism responsible for the appearance in the periphery of mature CD8 T cells bearing #autoreactive #TCRs that are absent from the thymus" www.science.org/doi/10.1126/...

After a recent prognosis given for #thymic #cancer that I basically have ten years left, I've decided to do something meaningful. I'm now helping to look after these gorgeous little chaps and chappesses before their eventual release into the wild. This is a dream. 🦔❤️💕
