These results suggest that temporally controlled SUMO pathway inhibition could be a novel strategy to modulate adipose tissue thermogenic capacity.
Key findings include:
· Transient TAK-981 exposure leads to robust and stable induction of beiging genes, including UCP1, and increased mitochondrial respiration.
· Mechanistic insights reveal immediate chromatin and enhancer activity remodeling and mobilization of CEBP family members and PPARG.
We investigated how the SUMOylation pathway regulates adipocyte plasticity. We discovered that brief pharmacologic inhibition of SUMOylation primes human pre-adipocytes for stable de novo beige differentiation when combined with PPARG activation.

IRX3 buffers the SUMOylation to promote adipogenesis and block osteogenesis. IRX3 loss boosts SUMOylation, inhibits PPARγ, and pushes cells toward bone fate, which is reversible with SUMOylation inhibition.
IRX3 is an epigenetic gatekeeper.

I am seeking a Pos-doc Researcher in Molecular Biology and Epigenetics. Join our team @biovitenskap.bsky.social @uio.no to explore how SUMOylation regulates epigenetic fate, gene expression, 3D-genome and chromatin during adipogenesis, with implications for obesity and cancer research.
This study is a very elegant characterisation of the epigenetic and transcriptional divergence between white and beige adipocytes. Very useful to those studying the epigenetic mechanisms supporting cellular reprogramming.

Our recent in vivo study reveals that inhibiting SUMOylation with TAK-981 disrupts fat accumulation, leading to impaired adipogenesis and increased immune cell infiltration in perigonadal white adipose tissue.

The project involves integration of ATAC-seq, ChIP-seq, CAGE and HiC experiments to reveal mechanisms that prevent epigenetic and transcriptional drifting during cellular reprogramming with a particular focus on the adipose tissue.
