These results suggest that temporally controlled SUMO pathway inhibition could be a novel strategy to modulate adipose tissue thermogenic capacity.

· Transient TAK-981 exposure leads to robust and stable induction of beiging genes, including UCP1, and increased mitochondrial respiration.

We investigated how the SUMOylation pathway regulates adipocyte plasticity. We discovered that brief pharmacologic inhibition of SUMOylation primes human pre-adipocytes for stable de novo beige differentiation when combined with PPARG activation.

IRX3 buffers the SUMOylation to promote adipogenesis and block osteogenesis. IRX3 loss boosts SUMOylation, inhibits PPARγ, and pushes cells toward bone fate, which is reversible with SUMOylation inhibition.
IRX3 is an epigenetic gatekeeper.
