The transcriptional repressor Fli1 inhibits proteostasis during nutrient stress to limit NK cell persistence in solid tumors Tumor-infiltrating NK cells are often dysfunctional. Ji et al. show that the solid tumor microenvironment metabolome drives proteostasis imbalance and dysfunction in tumor-infiltrating NK cells and demonstrate that deletion of Fli1, a transcriptional repressor of the unfolded protein response, enhances proteostasis and NK cell persistence, resulting in improved antitumor responses in vivo.
