IL-1β+ macrophages and the control of pathogenic inflammation in cancer While highlighting the complexity and heterogeneity of tumor immune microenvironments, the application of single-cell analyses in human cancers has identified recurrent subsets of tumor-associated macrophages (TAMs). Among these, interleukin (IL)-1β+ TAMs – cells with high levels of expression of inflammatory response and tissue repair genes, but with limited capacity to stimulate cytotoxic immunity – are emerging as key drivers of pathogenic inflammation in cancer. In this review we discuss recent literature defining the phenotypical, molecular, and functional properties of IL-1β+ TAMs, as well as their temporal dynamics and spatial organization. Elucidating the biology of these cells across tumor initiation, progression, metastasis, and therapy could inform the design and interpretation of clinical trials targeting IL-1β and/or other inflammatory factors in cancer immunotherapy.

TREM2 depletion in pancreatic cancer elicits pathogenic inflammation and accelerates tumor progression via enriching IL-1β+ macrophages Pancreatic ductal adenocarcinoma (PDAC) has a complex tumor microenvironment enriched with tumor-associated macrophages. Triggering receptor expressed on myeloid cells 2 (TREM2) is highly expressed by...

More on the role of IL-1 inflammation in TME... Deletion of TREM2 in pancreatic cancer unexpectedly accelerates disease progression due to secondary increase of pathogenic IL-1b+ TAMs 🔥🔥🔥

Type III interferons induce pyroptosis in gut epithelial cells and impair mucosal repair Intestinal damage following colitis or irradiation induces type III interferons (IFNs), which delay the healing of the intestinal epithelium. IFN-λ directs the sensing of Z-form nucleic acids generated during gut injury and repair, triggering cell death and altering healthy epithelial regeneration.

#EveryCellIsAnImmuneCell!💥Thrilled for the final version of our paper @cp-cell.bsky.social! We show that type III #interferons control ZBP1 activation driving gasdermin C cleavage & delaying gut repair by inducing #pyroptosis in intestinal epithelial cells! 1/n www.cell.com/cell/abstrac...
