Identification of Natural-Product Inhibitors of the 2C-Methyl-d-erythritol 4-Phosphate Pathway To tackle the emerging resistance against existing antibiotics, we screened natural-product (NP) libraries against two underexploited target enzymes from the 2C-methyl-d-erythritol 4-phosphate (MEP) pathway, namely, Mycobacterium tuberculosis DXPS and Escherichia coli IspD. We have chosen these two enzymes due to the availability of the crystal structures that helped to elucidate the putative binding modes of the NPs identified. The screening of a NP collection led to the discovery of myxobacteria-derived maracen A and Streptomyces-derived polyketomycin as the first NPs targeting these enzymes.

Our #DZIF natural product library was recently screened against two target enzymes from the MEP pathway. This led to the discovery of the first natural compounds targeting these enzymes. The results were published in ACS Med Chem Lett. Congrats to all co-authors!

🔥 Breakthrough in TB research! The antibiotic BTZ-043 reaches hard-to-treat granulomas and fights bacteria even in necrotic tissue. This is a major step toward more effective TB therapy! 🚀🦠

AT the #DZIF 2025 conference, we just had the privilege to listen to STewart Cole from Institute @pasteur.fr talking about Antimicrobial resistance #AMR. This is a massive risk for our society, but the financial incentives are skewed and most pharma has been divesting from antibiotic development.🧪
