Gotta share the negative results too! Looks like first round of copper tests don't show any signs of blue color from our #BlueSoup azurin expression plasmid in BL21. Media is slightly blue due to CuSO4 1mM addition (right). Spindown pellet is identical to no copper flask (left). Will let it shake...
I split a single PCR-validated colony of BL21 ecoli carrying a homebrewed expression vector containing a putative azurin from our beloved #BlueSoup into two volumes of 25mL LB-Amp. Added 1mM CuSO4 to one flask. Shaking at room temp (~22°C) and 200rpm overnight. Let's see if blue tomorrow🤞

I got a lil space in my docket today so I'm revisiting the Azurin transcription unit I built using the #BlueSoup sequence data. Gonna try to transform it into BL21 cells and grow on LB with copper ions in the media.

#BlueSoup azurin expression vector assembled and transformed. Let's see what we get tomorrow. I'll patch positive colonies onto LB-Amp with some CuSO4 (few mM) to help the azurins bind the ions and blue up. Will sequence and retransform into BL21 to boost expression. Exciting!
