HLA export by melanoma cells decoys cytotoxic T cells to promote immune evasion Melanoma cells escape immune surveillance by releasing MHC-antigen-loaded large EVs, known as melanosomes, that directly engage and impair CD8+ T cell receptors.

#WeekendRead! #SmartTumor #EveryCellIsAnImmuneCell! Levy & (many) friends show @cp-cell.bsky.social that melanoma tumors release large extracellular vesicles #melanosomes decorated with MHC-I & high affinity tumor associated antigens which subvert cytotoxic T cells! www.cell.com/cell/fulltex...

Breaking the mold: Unconventional T cells in cancer therapy Unconventional T cells, including invariant natural killer T (iNKT) cells, gamma delta (γδ) T cells, and mucosal-associated invariant T (MAIT) cells, play important roles in both innate and adaptive immunity. These cells respond to tumors rapidly and influence the tumor microenvironment (TME). Recent advances in understanding their biology, as well as the development of novel therapeutic approaches, have underscored their potential in cancer immunotherapy. This commentary will assess these advances and translational possibilities in the field.

The HIP mouse and all of its organs are completely invisible to allogeneic immune cells Hypoimmune (HIP) allogeneic cell therapeutics hold the promise to allow off-the-shelf treatments for a broad patient population. Our HIP approach includes the depletion of MHC class I and II molecules and the overexpression of Cd47. Here, we report the engineering of HIP mice that stably exhibit the HIP phenotype in all cell types. Parabiosis experiments were designed to broadly assess immune evasiveness of all HIP blood cells in fully allogeneic BALB/c mice. HIP blood cells did not induce any immune response and achieved stable engraftment in BALB/c mice.

Generation of T cells with reduced off-target cross-reactivities by engineering co-signalling receptors

Our new work!

We show that T cell cross-reactivity can be reduced without modifying the TCR itself (by dramatically enhancing antigen discrimination).
