Oh wow.

Some fraction of AMPAR heteromers incorporate CNIH1-4 in HEK cells. That was always my fear about switching to *another* cell line- we would not know the mixture.

Calcium and magnesium in the pore of NMDA receptors!

Lipids play an unexpected role. The hydration shells of Ca2+ and Mg2+ could be the difference between permeation and block. So much physiology in the energetics of H2O.

Nice to see these detailed views of my dear GluK2, and the progress in the last 20 years since the first structures.

But perhaps the most important experiment to do now is the single channel recording of the staircase activation, to see if subconductances are there.

Activation of kainate receptor GluK2–Neto2 complex - Nature Structural & Molecular Biology Gangwar et al. present cryo-electron microscopy structures of GluK2 kainate receptors with auxiliary subunit Neto2 and reveal the basis of receptor activation.

Kainate receptor modulation by Neto including an open state. Surprising, to me: I would never have imagined that Neto works like a membrane-tethered lectin (ConA). Auxiliary proteins influence dimer-dimer interactions, as we and others also showed in AMPARs.

AMPAR inhibition lowers S1P in human blood and improves murine autoimmune neuroinflammation by blocking T cell egress from lymph nodes A clinically available AMPAR antagonist inhibits T cell egress from lymph nodes and shows therapeutic effects in mouse models of multiple sclerosis.

Interesting results. Perampanel (AMPA blocker approved for epilepsy) has a big effect on T-cell mobilisation.

Perhaps an off-target effect? Maybe I missed a KO control using the floxed GluA123 mouse.

But useful either way, given that Pmp is quite mild.

Nice paper. I recommend to check the review file for a lot of extra interesting details. Still a lot to understand about TARP action, despite >20 years of work from many groups.

You want to see kainate and forget AMPA? GYKI 53655 is still your best bet.

Unfortunately if you use a lot, you will also start to hit some kainate. It's not terrifically selective. But at 30 µM you should be fine.
