To identify novel regulators of the #MDA5 pathway, we used SILAC-MS in #virus infected vs uninfected cells. We identified ~100 MDA5 binding partners, one of which was Annexin A2 (ANXA2).

This dsRNA is likely from aberrant transcription of non-exonic RNA in/near ISGs, often introns that contain repetitive elements - sequences known to form double-stranded structures that activate dsRNA sensors like #MDA5 and #PKR.

Such a tour de force paper, a really nice read!

Wow what an amazing article! Very cool to see the important role of #macrophages and #interferon in the #tumour microenvironment. I would bet 💵 that the IFN-driving azacitidine works via #MDA5, as this has been shown to activate MDA5 via ERV expression in cancer cells 😄!

Through TLR3, ZIKV induces proinflammatory #cytokines (IL-6, IL-10), which trigger STAT3 signaling, which in turn dampen #interferon responses. As #RIGI and #MDA5 sense ZIKV and induce IFN production, this antagonism eventually aids viral replication as we can show.
