File systems are often invisible, but so important! #relion may run multiple times faster on a well tuned file system, compared to a bad one. Appreciate your local compute people who set this up for you, and be patient with them when it fails (which is very stressful).

These results are truly impressive! But I do worry that #cryoEM is moving into a phase where progress is made by optimizing parameter settings for black box programs. The devil is often in the details, and I am not sure how this could be implemented in #relion, as stated, without more info.

At its caps, there is no space for 39 MVPs. Using symmetry expansion and partial signal subtraction in #RELION, we show that the symmetry at the caps is broken and 2 out of every 3 MVP monomers gets successively excluded from the cap.

If it helps anyone receiving a rejection today: my original paper on #RELION was rejected by @pnas.org and Structure @cellpressnews.bsky.social. The method still did pretty well.

Please note that this is not new: it basically is Bayesian Polishing from #RELION. Academics invent these algorithms, which get picked up by commercial companies. This is great, as #OpenSoftwareAcceleratesScience, but don't forget where it all came from.

I should also thank @SjorsScheres and other #Relion developers! 😃 I racked up a whopping ~700 jobs trying to get the most out of this data (but I'm sure someone can improve the resolution). The biggest improvement came from increasing the box size 😳
