Mapping cis -regulatory mutations at scale in sorghum enables modulation of gene expression To understand how to improve photosynthetic gene expression using precise, small CRISPR-type edits (Fig. 1a), we developed an MPRA in sorghum protoplasts to identify hypermorphic and hypomorphic mutations within crDNA (Fig. 1b). Briefly, tens of millions of healthy mesophyll protoplasts were isolated from partially etiolated Sorghum bicolor cv. RTx430 seedlings (Extended Data Fig. 1 and Methods) and transfected with plasmid libraries containing crDNA variants. After an 18-h incubation in darkness, cells were exposed to 4 h of light to stimulate photosynthetic gene expression. Next, barcoded mRNA was harvested, next-generation sequencing was used to quantify barcodes in the input DNA and output RNA and linear regression was performed to infer individual variant effects, Emut (Methods and Extended Data Fig. 2). Viruses That Jump to Humans Don't Need Special Mutations, Study Finds - Science Alert When a virus jumps to humans from another species, it can seem like a supervillain bent on world domination – especially if it goes on to ignite a pandemic, as many zoonotic viruses have done in recent decades. And yet, despite the genuine danger zoonotic pathogens pose to human civilization, they're often just capitalizing on conditions we created, like our proximity to livestock, our degradation and invasion of wildlife habitats, and our trading of captive wildlife. In fact, according to a new study, most of the zoonotic viruses behind recent epidemics or pandemics were surprisingly unremarkable before they jumped to humans, with no clear evidence of prior adaptive changes predisposing them for such a leap. This challenges a common assumption about spillover events, which have long been seen as breakthroughs in which viruses leverage a novel adaptation. Instead, the new findings suggest our spillover risk may be even greater than we thought. If spillover depends less on sudden pathogen mutations than on our encountering existing ones, we're sitting ducks. This suggests that the D108N mutation introduced in the first round of directed evolution of ABE7.10 was critical for both enhancing DNA editing activity by TadA and maintaining this activity even in the context of the 13 other acquired mutations. To investigate how the additional mutations in ABE7.10 impact base editing activity, we generated all 14 single-residue ABE7.10 reversion editors and evaluated their activities using a previously reported set of 8 episomal, highly sensitive, fluorescence turn-on reporters (Fig. 2a and Supplementary Discussion 1)12.
