Protocol to generate human monocyte-derived macrophages in the absence of exogenous proteases and protease inhibitors to study proteolysis Investigating proteases and protease inhibitors produced and secreted by primary cells is complicated by the presence of exogenous ones in cell culture medium containing fetal bovine serum. With this protocol, we generate human monocyte-derived macrophages in the absence of exogenous proteases and protease inhibitors by using a serum replacement for the final 24 h of macrophage culture. We describe steps for seeding and differentiating monocytes into macrophages. We then detail procedures for ma...

Not long after I started my lab, a prominent scientist told me it was β€œimpossible” to be a successful PI and a mother

Don’t ever let the naysayers tell you it can’t be done - of course it can!

#Updated program

Unfortunately, John McKendrick from UGent cannot present at #yBIS2025.
Jennifer Vandooren ( @kuleuvenuniversity.bsky.social) will share her research on the 9th of May on leukocyte proteases as immune mediators.

But what changed compared to the original preprint?

Also, I feel i should post Movie 1 πŸŽ₯, that inspired the cover. Back when I did the original bluesky thread, movies were not available.

microscope image of a postnatal mouse brain showing the entry point of blood vessel into the brain - fibroblasts (yellow: S100a6) form a layer at the brain's edge (meninges) and surround the blood vessel. Astrocyte endfeet (magenta, GFAP) form a layer at the brain's edge, apposed to the fibroblasts. Image also shows macrophages (cyan; CD206) and cell nuclei (DAPI, blue)

I was imaging this weekend, took this image because it was beautiful to me, the blood vessels, fibroblasts, astrocytes, & macrophages as steadfast guardians at the brain's edges. Cheers to the 'guardians' of all that is important. ❀️

Human monocyte-derived macrophages shift subcellular metalloprotease activity depending on their activation state Natural sciences; Biological sciences; Immunology; Immune response

Check out our latest research πŸ‘‡πŸ§ͺπŸ”¬πŸ‘©πŸ»β€πŸ”¬! We studied how protease activity changes in human macrophages upon their activation. We uncovered that IL-4 results in the uptake of matrix metalloprotease-9 (MMP-9) and its nuclear localization. #Macrophages #Immunology #AcademicSky
